Proteomic dysregulation was investigated in a human T-cell lymphoma model to explore IL-17A-mediated pathogenesis. Label-free quantification was employed to determine fold changes in protein abundance between IL-17A-treated and untreated HuT 78 cells. Specifically, HuT 78 cells were treated with cytokine IL-17A (100 ng/ml), followed by protein extraction using the Trizol-chloroform method. The extracted proteins were digested with 1 microgram of MS-grade trypsin, and the resulting peptide samples were injected into a Thermo Scientific Q-Exactive Plus mass spectrometer, coupled with an EasyNLC HPLC autosampler and C18 PepMap trap column. Mass spectra were analyzed using the Thermo Scientific Proteome Discoverer platform (version 2.2), employing Mascot and SequestHT as search engines. Standard workflows for discovery proteomics were applied. This dataset successfully modeled IL-17A-driven inflammation in HuT 78 cells, revealing numerous dysregulated signaling pathways, which may provide potential targets for novel therapeutic strategies
